This report ‘Anticancer Therapeutics’ provides a comprehensive overview of the wealth of information now available in this important and fast-moving subject. The book provides a clear introduction to the area, with a historical overview of initial developments of cancer therapeutics and their progress in today’s multidisciplinary approach to cancer treatment. The book, written by the winner of the 2008 Mike Price Fellowship, Dr Sotiris Missailidis of Department of Chemistry and Analytical Sciences at the Open University, presents the current therapeutic regimes, with their indications and side effects, as well as their position in the international market in terms of sales and development costs. Furthermore, the book includes a discussion of our advancement in the understanding of cancer biology and how this has driven the drug discovery process. Modern drug discovery aspects, through genomic, proteomic and metabolomic approaches are referred to as well as combinatorial chemistry techniques and discovery of chemotherapeutic agents from plant extracts, re-use of old drugs and drugs from other indications, or de novo rational drug design. These have lead to the basic areas of drug development, such as chemotherapy, immunotherapy, radiotherapy, gene therapy, cancer vaccines and potential experimental therapies that may influence the cancer scene in the next 10-20 years, including aptamers, antisense and antigene, siRNA etc.
The book provides an opportunity to compare such novel therapies with small molecules and biologics, such as Gleevec and Herceptin. Finally, other crucial aspects in current and emerging anticancer therapeutics, such as development of resistance to chemotherapy, including the various processes that drive it, and the pharmacological aspects of cancer therapy will be discussed.
