Special Report: The Autoimmune Biologics Pipeline-A Competitive Review
Decision Resources, Inc, Aug 2011
Although biologics such as the tumor necrosis factor-alpha (TNF-alpha) inhibitors have vastly improved the treatment of various autoimmune disorders, many patients are refractory to these and other therapies. Treatment of these patients represents an opportunity for developers of biologics to discover and develop biologics with different targets. For example, increased levels of interleukin-6 (IL-6) have been found in patients with rheumatoid arthritis (RA) and Crohn's disease (CD), making this cytokine an attractive drug target for biologics (e.g., Roche/Chugai's Actemra/RoActemra [tocilizumab]) to treat autoimmune disorders. With a pipeline of 74 agents (vetted from an initial list of 445 agents, including small-molecule drugs), drug developers are working toward marketing approval of autoimmune biologics with novel and existing targets. - Agents in the autoimmune biologics pipeline aim for a wide array of drug targets. What are the most common drug targets? Why are drug manufacturers aiming for these targets instead of others?
- The autoimmune biologics pipeline focuses on a diverse selection of indications. What are the most commonly targeted autoimmune indications?
- Despite the success of the TNF-alpha inhibitors and other biologics, significant unmet needs remain for many autoimmune indications. What autoimmune indications could benefit most from newer biologics? What unmet needs are most striking in these autoimmune indications?
