In turn, the DNA breaks stimulate the cell’s endogenous repair mechanisms, which are typically non homologous end joining(NHEJ) or homology-directed repair (HDR), depending on the cell state or the presence of a repair template.

What are the Major Genome Editing Technologies?

The currently available genome editing technologies encompass several classes of programmable nucleases, including mononucleases,zinc finger nucleases (ZFN), transcription activator-like effector nucleases(TALENs), and CRISPR(clustered regularly interspaced short palindromicrepeats)-Cas9 nuclease systems. ZFNs, TALENs and CRISPR-Cas9 are the most used and commercially developed technologies at the present time.

While NHEJ is active throughout the cell cycle, HDR occurs only in dividing cells.  NHEJ repairs the DNA by directly rejoining the two cut ends; however, as NHEJ is an error-prone process, it can introduce insertion or deletion (indel) mutations in the genome that can lead to the degradation of mRNA or the production of truncated proteins,ultimately inactivating the function of that particular gene. On the other hand, in HDR, the lesion is repaired by inserting an exogenous DNA template atthe DNA break location, thus replacing a mutated gene or introducing new genes of interest.  The currently available genome editing technologies encompass several classes of programmable nucleases, including mononucleases, zinc finger nucleases (ZFN), transcription activator-like effector nucleases (TALENs), and CRISPR(clustered regularly interspaced short palindromic repeats)-Cas9 nuclease systems. ZFNs and TALENs are naturally occurring chimeric enzymes comprising a DNA binding domain fused to a non-specific FokI nuclease domain with DNA-cleaving activity.
