Bruton tyrosine kinase (BTK) is a nonreceptor tyrosine kinase which plays a central role in the signal transduction of the B cell antigen receptor (BCR) and other cell surface receptors, both in normal and malignant B lymphocytes. Long-term use of BTKi, however, is expensive and can be complicated by the emergence of long-term toxicities and/or resistance. Second generation kinase inhibitors may have an improved side-effect profile that may facilitate long-term use, while combination therapy approaches induce deeper remissions which opens the door to limited-duration therapy.

It is approved for the treatment of mantle cell lymphoma, chronic lymphocytic leukemia or small lymphocytic lymphoma and Waldenström's macroglobulinemia.
