A commonly measured parameter in the cell viability assays is the integrity of the membrane. Cell-impermeant is membrane integrity dyes that can only enter live cells when there is a compromised plasma membrane. Usually, the membrane integrity viability dyes are made of Nucleic acid binding dyes and amine-reactive dyes.

A nucleic acid binding dye is nonfluorescent in aqueous media but shows increased fluorescence after joining with a double standard DNA or RNA. Propidium iodide (PI), SYTOX dead cell stains, and 7 amino actinomycin D (7 AAD) are some nucleic acid viability dyes. Amine reactive dyes bind covalently with the primary amine group available on the cellular proteins, which are present on the interior and exterior of the cells.

The signal is weak when the amine proteins are attached to the surface but showcases high signal intensity when the dye penetrates the cell with a compromised plasma membrane and reacts with the interior amine cellular protein. For flow cytometry, amine-reactive viability dyes contain live/dead fixable dead cell stains.
