ATP (Adenosine triphosphate) cell viability is an assay used to measure cell viability since only viable cells can make ATP. Reagents with the CellTiter-Glo luminescent cell viability assay that contains detergent, luciferin substrate, and stabilized luciferase can be used to measure ATP. The detergent lyses viable cells, discharging ATP into the medium.

Luciferin is used by luciferase to create luminescence in the presence of ATP, which can be detected in a few minutes using a luminometer. The CellTiter-Glo 2.0 Assay is given as a single solution that decreases the reagent preparation time and allows storing it at room temperature, making implementation easier.

As these ATP assays don’t require lengthy incubation times to convert a substrate into a colored product, they usually work faster than other methods. In addition, they have wide linearity and excellent sensitivity, making them very suitable for high throughput applications with low cell numbers. They also have reduced risks of artifacts than other methods.
